1. Overview
Cagrilintide is an amylin analog developed by Novo Nordisk for obesity treatment.
Unlike GLP-1 drugs (e.g., semaglutide, tirzepatide), it works via amylin receptor agonism, reducing appetite and slowing gastric emptying.
15mg dose is not yet studied in humans-highest tested dose in trials was 4.5mg weekly.
2. Clinical Trial Data (Existing Doses)
Phase 2 Trials:
1.2mg, 2.4mg, 4.5mg weekly were tested.
4.5mg showed ~10-12% weight loss over 26 weeks (vs. placebo).
Combination with Semaglutide (CagriSema):
2.4mg Cagrilintide + 2.4mg Semaglutide → ~15-17% weight loss (superior to semaglutide alone).
3. Is 15mg Cagrilintide Being Studied?
No public data supports a 15mg dose.
Novo Nordisk may explore higher doses in future trials, but current focus is on CagriSema (lower-dose combo).
4. Safety Concerns with High Doses
Nausea/vomiting is dose-limiting (already significant at 4.5mg).
Risk of pancreatitis or hypoglycemia could increase at 15mg (theoretical risk).
